Missing Link: Neurotransmitter Lithium

In spite of your original article on the study "of cross-reactivity among proton-pump inhibitors", Allergy 2005; 60; 1087-1093, and the AllergyNet article "a new role of TSH in immune response", Allergy 2005; 60; 489-493, I like to remember to the work done by my groups (1-3, 5): 1. Substituted immidazoles are disturbing dithiol/disulfide-balances/shifting redox-potentials and/or collapse potassium(sodium) gradients, established actually by potassium(sodium)/water pumps and not by "proton pumps". Which explains the allergic reactions among the "proton-pump inhibitors". 2. The improvement of the immune state by a thyroid hormonal therapy is to explain by "the missing link" neurotransmitter Lithium, which modulates L-thyroxine reactions....and not "new role" TSH hormone! The reaction mechanism of the proton substitute metal lithium is given for convenience:
Lithium has a myriad of effects on cellular transport mechanisms, second messenger pathways, and neurotransmitter dynamics. The two major involved neurotransmitters in the pathogenesis of affective disorders are 5HT and NE, which are modulated by Li. Li-salts are useful in preventing migraine attacks and shortening manic periods. Li-treated bipolar subjects show lowered brain to serum concentrations (7Li NMR): Li disturbes H-bridges and the electric conductivity, slows down oscillations involved in memory and thinking, in thermoregulation. Li injections into the frog skin gives rise to electrical oscillations.
Li decreases noradrenergic transmission, NE release, inhibits basal and beta adrenoceptor-dependent adenylate cyclase, modulates the phosphatidylinositol pathway, interacts with acetylcholine/ligand-binding and its induced flux, modulates ligand- and voltage-gated ion channels. Li is an ion that substitutes for sodium and/or protons: Li distributes rapidly to liver and kidney, Li enters cells via sodium channels, incl. K(Na)/H2O-pump (F0F1-ATPase): extruded by the active cation pump (and e--driven K,Na/H-exchange-system) at about one-tenth of the rate of sodium: Li accumulates therefore in cells and modulates cellular volume. Li interferes with cation binding, with membrane fluidity by interfering with phospholipids/lipids. nAcChR and K/H2O-Pump were similar in many aspects.
Li inhibits Na/K-ATPase, gives rise to hypertension by induction of (erythrocyte) Na/Li countertransport, Li/Na-exchange-activity; results in hyperthyrodism (inhibition of jodine uptake), in progressive chronic nephropathy, in hyperthermia.
Secondary effects of Li were its anti-inflammatory activity, its immunostimulating and antimirobial properties: its potentiation or suppression of immunity, its possession of pro- and anticancer properties (2-5). Li is found to be essential for our body: Li itself seems to be a neurotransmitter and/or regulator of their action.
The side effects of Li includes fatique. Above 2 mEq/L more severe toxic signs may occur. The toxicity of Li is increased by factors that deplete the body of sodium. Restriction of dietary sodium dramatically increases the renal reabsorption of Li and can lead to severe toxicity. Similarly, diuretics that lead to sodium loss augment Li toxicity: kidney damage, chronic allergic reactions.
Lithium is not metabolized, and elimination is predominantly via filtration in the kidney. Renal impairment results in accumulation of Li, with resulting toxicity.

Thank for this list which acknowledges the impact you have in your field.
However, the three references you quoted in your letter were not (to my
knowledge) easily accessible on Medline or EMBASE. Thus, the authors of the
paper could not mention them.
I have read carefully the letter, and due to its interest and the lack of
space in our Journal, I decided not to publish it.
